Since December last year we've had deepdewedge, icecream, and now isonet 2 - its probably time a really thorough review of image restoration in cryo-ET, but at this rate it will be obsolete before it comes out!

I'd also say that I'm the first to say quantification and statistics are critical for cell biology, but also seeing is believing and generating better visualizations is critical for the continuing broad value of our field!

I'm curious if you have a good way to measure that! We have found that basically all the tools are near-perfect at enhancing segmentation, but aren't great for template matching. Improvements tend to be in training and avoiding artifacts, which I'm not sure how to compare.

Identify particles that can be averaged in the raw data, average them, then compare the average reconstruction with denoised/dewedged data, preferably per-Fourier-space-voxel rather than directional FSC to figure out what happens in the missing wedges

I don’t n know, but when I see all the scientific and artistic advances that humans are capable of, day in day out, why for heaven’s sake are we wasting time and resources making wars and being bitter and hateful to one another? There has to be something deeply rotten in the human psyche
