nature.comA CD4-lineage erythro-myeloid progenitor within the adult bone marrow defines an alternative pathway for macrophage generationWe have previously described a distinct population of tissue-resident macrophages termed PVMs, which localize to the perivascular space of postcapillary venules in the dermis25. As part of the perivascular extravasation unit, PVMs mediate the recruitment of leukocytes into the skin during inflammation26. The precise ontogeny of PVMs remains controversial. The selective identification of PVMs is greatly facilitated in the transgenic mouse strain DPE-GFP27, in which regulatory elements of the Cd4 locus drive the expression of green fluorescent protein (GFP25; Supplementary Fig. 1a). Besides T cells and plasmacytoid dendritic cells (pDCs), PVMs, but not interstitial macrophages, selectively express GFP in these mice. In addition to the dermis25, CD11b+CD64+F4/80+GFP+ PVMs are present in other organs, including the mammary fat pad, the meninges and the superficial layer of central nervous system (CNS) cortex, in DPE-GFP and DPE-GFP:Rag1−/− mice (Fig. 1a and Extended Data Fig. 1a). Similar to the skin, confocal microscopy of inguinal mammary fat pads revealed that GFP+ cells lined the blood vessels (Fig. 1b, left). Co-staining with anti-F4/80 confirmed the identity of these cells to be macrophages (Fig. 1b, right). Intravital multiphoton microscopy illustrated that these cells were sessile with dendrites that actively probed the surrounding tissue microenvironment (Fig. 1c and...
