New research. Conveniently, Marchantia has relatively few transcription factors. Yet, we need to leverage on that at the family scale to have a more comprehensive picture. In this paper, we focused on ERFs, the second largest TF family. #PlantScience www.biorxiv.org/content/10.6...

Interestingly, we found that the MpERF2 makes ectopic rhizoids. A surprising function compared with its Arabidopsis WIND homologues. Also, it is a proof-of-concept for doing functional genomics at scale. I hope the #evodevo community enjoys it, and feedback is very welcome.

We made gain-of-function screening for practically all ERFs in Marchantia and found that most have very strong and distinct developmental phenotypes. Different to their classic function in flowering plants associated with stress and ethylene responses.

Delighted to share our paper on neo-chromosomes! We induced centromere fission using CRISPR in Arabidopsis and increased the chromosome count from n=5 to n=6. The broken chromosomes ends quickly “healed” by adding telomeres de novo and the new karyotype is stable through mitosis and meiosis! 1/3

Excited to share this story in its final form, now published in ACS Synthetic Biology. #PlantBiology #Synbio pubs.acs.org/doi/10.1021/...

pubs.acs.orgEngineering Auronidin Production in Marchantia polymorpha Enables a New Class of Plant-Derived Textile PigmentsPlant-derived pigments offer sustainable alternatives to synthetic colorants, yet their practical deployment in textiles is limited by restricted chemical diversity and low abundance. Liverworts represent a source of diverse chemical compounds, and the model liverworts Marchantia polymorpha are emerging as chassis for bioengineering and synthetic biology. Here, we report the biotechnological application of auronidins, a rare class of flavonoid pigments, as textile dyes. Using M. polymorpha, we engineered enhanced auronidin production through controlled expression of the R2R3-MYB transcription factor MpMYB14. We systematically benchmarked constitutive and inducible gene expression systems, including heat-shock, glucocorticoid receptor, and β-estradiol (XVE) circuits, identifying inducible strategies that decouple biomass accumulation from secondary metabolite production while achieving high pigment yields. Extracted auronidins were used to dye cotton yarn directly, demonstrating the feasibility of auronidins for textile dyeing. Our results establish M. polymorpha as a versatile plant chassis for programmable secondary metabolite production and introduce auronidins as a promising natural pigment platform for sustainable textile biotechnology.

Marchantia might seem an unlikely biotech crop, but it’s worth trying. In Aurore’s thesis, we pushed this concept by engineering the auronidins with MpMYB14 and using the pigments to dye cotton. It ended up giving a very unique and beautiful product. #PlantScience www.biorxiv.org/content/10.6...

Our first study on how the leaf succulent Kalanchoë laxiflora makes stomatal subsidiary cells is finally peer-reviewed & out - and it made the cover! 🌵 🧬 🔬 doi.org/10.1126/scia... Despite 150 years of evolution, grasses and Crassulacean succulents use the SAME gene to make subsidiary cells! 🤯

🌵Preprint alert 🌵 5y in the works & extremely proud of this one. Xin Cheng & @heikelindner.bsky.social established the eudicot leaf succulent Kalanchoë laxiflora as a dev model. Like in grasses, MUTE makes Kalanchoë’s stomatal subsidiary cells! 1/n www.biorxiv.org/content/10.1... BSKY-tutorial 👇
