We propose: Synapses donโ€™t lift inhibition by physically removing SynGAP1. They do it by: โ€ข functionally inactivating existing SynGAP1 โ€ข halting its local production โ†’ Translational repression acts as a โ€œplasticity gateโ€ controlling when synapses can strengthen. 6/7

Despite abundant dendritic Syngap1 mRNA, translation isnโ€™t constant across time. Instead, SynGAP1 synthesis is dynamically repressed: โ€ข during synaptic maturation โ€ข and crucially, transiently suppressed after LTP induction 4/7

We also challenge a key model: SynGAP1 does NOT disperse from synapses during LTP. That effect comes from GFP overexpression artefacts, as the endogenous protein remains stably anchored at the PSD. 5/7

Most studies on locally translated synaptic proteins focus on those that promote strengthening. But what about the brakes? We looked at SynGAP1 โ€” a major negative regulator of Ras/Rap signalling โ€” to ask if it is locally made to constrain plasticity? 2/7

We find SynGAP1 isoforms are tightly developmentally regulated: โ€ข ฮฑ1 = stable synaptic occupancy โ€ข ฮฑ2 = progressively enriched (dominant at synapse) โ€ข ฮฒ = accumulates later in dendrites All isoforms sit right at the PSD. 3/7

Spent 10k on custom media from @thermofishersci.bsky.social which kills our neurons, unlike a similar in house made version. Complained to thermo about this, and after a month of โ€œinvestigationโ€ they refuse to refund or replace it- but will let us have a discount on a replacement. Well done.

On Wednesday I had to say goodbye to my loving George after 13 years. He was sweet, needy, dumb as a rock, and an all around amazing loaf of orange fur. Iโ€™m going to miss this bestest dude.
