We are a lab studying the molecular mechanisms of epigenetics and how epigenetics impacts a variety of biological processes and diseases.

"We are living with the legacy of the McCarthy era, and what Donald Trump is doing is really resurrecting the worst."
After a performance of "Are You Now Or Have You Ever Been," @dlnyclu.bsky.social spoke at a talkback on the attacks we're enduring on free speech and the fight for our democracy.
Very excited about our recent preprint- @lindashuyingxu.bsky.social and collabs discover that neutrophils repurpose their nucleolus for storage of pre-made IFN-I to be rapidly released via a nucleolar stress response that results in nucleolar extrusion and secretion.

Absolutely beautiful paper by a core team at the RIKEN in which the telomerase RNA component has (finally) been identified, in the nematode *C. elegans*. Surprisingly, it does not have its own promoter, but is located inside the intron of a germline-expressed gene (nmy-2)! 🤩

Really cool paper from Marie-Anne Felix's lab. Genetic variation in set-24 controls the duration of epigenetic inheritance.

Excited to share our new work on transgenerational adaptation www.science.org/doi/epdf/10.... out today spearheaded by outstanding former undergraduate @kathleenkim.bsky.social. C. elegans adapt to repeated generational stresses and no longer display altered fertility, fat content, + longevity 1/9
This exciting story was spearheaded by Kathleen but she was helped by a number of people mostly not on bluesky including Ariel Telger, Gautam Sarkar, Sharan Surya, Hafiz Rothi, @mpmeers.bsky.social, and Simon Yuan Wang. 9/9

We also performed a directed screen and identified that depletion of the argonautes CSR-1 and HRDE-1 accelerated the transgenerational adaptation (2 generations instead of 4 for no fertility phenotype) and depletion of the H3K27 trimethylase PRC2 complex eliminated the worms ability to adapt 7/9
Really excited for the next steps of this project to further study whether transgenerational adaptation is universal to different environmental stresses, how conserved this is in other species, and what the precise molecular mechanism is for the transgenerational adaptation. 8/9

We also tested elevated glucose which increases neutral lipids and decreases fertility in exposed worms and unexposed descendants. After two successive exposures to elevated glucose neutral lipid levels and fertility were the same as unexposed controls. Therefore transgenerational adaptation..5/9
..is not unique to hypoxia and might be a general phenomena. We performed RNAseq to examine which genes were adapting their expression to repeated generational hypoxia exposure. We found that most genes still respond to hypoxia treatment but a small subset adapt to repeated generational hypoxia 6/9

We examined the effects of exposing successive generations to a stress we knew would induce epigenetic inheritance. Hypoxia exposure causes extended lifespan, reduced fertility, and reduced lipids and some of these phenotypes are heritable pubmed.ncbi.nlm.nih.gov/36516753/ 3/9
We found that after 2 successive generational exposures to hypoxia, worms were no longer long lived. After 4 successive generational exposures to hypoxia, worms no longer displayed a reduction in fertility. The worms adapted on a transgenerational scale to the repeated environmental stress! 4/9

Recent work has demonstrated that a single exposure to a dramatic stress induces transgenerational phenotypes of altered color, obesity, glucose tolerance, behavior and longevity. But we are exposed to different environmental stresses each generation (changes in pesticides and fungicides etc) 2/9
MADELEINE DEAN: The president is unhinged. He's unwell
JOHNSON: A lot of folks on your side are too
D: Oh my god, please. That performance in front of the generals?
J: I didn't see it
D: It's so dangerous! Our allies are looking elsewhere. Our enemies are laughing. You have a POTUS who's unwell.
A federal judge in Boston said she found it "difficult to conclude anything other than that [the Trump administration] used antisemitism as a smokescreen for a targeted, ideologically-motivated assault on this country's premier universities, and did so in a way that runs afoul of [federal law]."
Donna Lieberman of @nyclu.org is absolutely right. It's vital that @governor.ny.gov sign these critical bills to protect New Yorkers' reproductive freedom, including the NY Health Information Privacy Act that I carry w/@lindabrosenthal.bsky.social.

We found that dimt-1 deficiency requires the TOR signaling pathway and an intact germline to extend lifespan. Using auxin-inducible degron tagged dimt-1 and tissue specific RNAi we found DIMT-1 functions in the worms' germline after mid-life to regulate organismal lifespan 5/8
We also found that DIMT-1 regulates selective translation of stress resistance and lifespan regulating transcripts in the worm's germline but not in the muscle. One transcript which we identified as displaying differential ribosome occupancy was the cytochrome P450 enzyme DAF-9. 6/8
We found that DIMT-1 functions in the germline and works after mid-life to extend lifespan. This reveals a new layer of regulation of lifespan, which transcripts are selectively translated, and found that ribosome heterogeneity is critical later in life to promote healthy aging. 3/8
An RNAi screen identified that dimt-1 KD caused lifespan extension. We had previously shown that DIMT-1 is an 18S rRNA m62A methylase and that methylated rRNAs are transmitted to subsequent generations to drive intergenerational response to starvation pubmed.ncbi.nlm.nih.gov/37689068/ 4/8
As we age there is a dysregulation of which proteins are degraded and which proteins are produced. We showed that DIMT-1 methylation of the ribosome selectively regulates the production of stress resistance and longevity regulating proteins and regulates aging. 2/8
I was born in the McCarthy era, but this is McCarthyism on steroids.
Our democracy is under dire threat. But we have a government of laws, not men. And NO ONE is above the law.
Across the country, the ACLU has already filed more than 50 lawsuits to defend free speech, due process, equal rights, and the rule of law.
We’ve got the assignment. And we’re not slowing down.

Last week, House Democrats opposed giving Elon Musk and Trump a blank check funding bill that would allow them to gut programs and services families rely on.
The Senate must pass a short-term spending bill so we can pass bipartisan bills that protect programs for our communities.
KEEP CALLING. PEEL OFF ALL DEM SENATORS YOU CAN.
☎️: (202) 224-3121
Tell them to vote NO on Cloture and NO on CR. Vote is pushed to after 1:15ish for now. Situation is fluid.
The only public YES votes are Schumer and Fetterman. They need 7-8 total. KEEP CALLING.
Call retiring Senators too!
CALLS ARE WORKING - it has bought us precious time, but we NEED to keep the pressure up.
Tomorrow is the cloture vote.
KEEP CALLING: (202) 224-3121 ☎️☎️☎️
Tell your Dem Senator to vote NO on Cloture and NO on the Republican spending bill.
Do NOT assume your Senator will be good on this. Trust me.
🚨 Everyone needs to call their Dem Senator right now. They are starting to cave.
Tell them:
1. Vote NO on Cloture
AND
2. Vote NO on the Republican spending bill.
Don’t let them pivot to reconciliation. GOP doesn’t need Dem votes on that and they know it.
TODAY is the showdown.
