We developed Adaptive Polishing for the consistent preparation of ~150 nm thin FIB-lamellae. This was done by using imaging feedback to adjust parameters for each lamella, or Adaptive Milling, rather than applying a fixed recipe. Preprint: doi.org/10.64898/202...

We also provide a direct comparison of the damage layers caused by milling with gallium, argon, and xenon at 30 kV, and find that xenon is significantly less damaging.

-Using a truncated ribosome, we predict the increased signal with Nilas would enable detection of 1.5x smaller macromolecules (220 kDa) -We find macromolecules from 2.5 MDa to 200 kDa within Nilas-milled lamellae, including glycolytic enzymes and RNA Pol III

We compare Nilas-milled lamellae to standard gallium 30 kV polished lamellae and show that Nilas produces: -Thinner lamellae with increased contrast -Significantly reduced damage layers -Higher signal and improved detection of ribosomal subunits -Improved 3D reconstructions with fewer particles
