Caribou Biosciences’ shutdown highlights the challenges facing allogeneic CAR-T therapies. Indeed, emerging in vivo CAR-T approaches generate autologous CAR-T cells directly within the patient, potentially at a fraction of the cost of conventional ex vivo manufacturing. buff.ly/MpQ0vOX #CART

Recent safety events with Novartis and BMS highlight the challenge of translating #CART from life-threatening cancers to chronic autoimmune diseases. Meanwhile, Kyverna’s miv-cel continues to show encouraging efficacy and safety in stiff person syndrome. BLA planned by year-end. buff.ly/TIe8ymE

Fate’s #ACR2026 studies suggest #iPSC-derived #CARTs could become a compelling allogeneic platform: a potentially unlimited supply of a standardized, homogeneous product. Data cover FT819 in lupus - without conditioning/outpatient use - and CD19/CD38 FT839 in RA. buff.ly/1U7ulvd

Early but promising results with armored #CARTcells targeting glypican-3 and co-expressing IL-15 and IL-21 in pediatric hepatoblastoma. Step by step, CAR-T cells are beginning to make solid tumors addressable. buff.ly/ZqJ8UbH

Craig Venter passed away. I'm old enough to remember the sequencing of the genome in 2001, by the team of Craig Venter (Celera Genomics), the Science cover, and an International public consortium, the Nature cover. buff.ly/InQ8izN

Two extraordinary charts from today’s PISA test results: 1) School test scores continue to collapse internationally, underscoring how this is no longer a Covid effect but sustained decline. Those falls in reading and maths are equivalent to about two years of lost schooling.

In a Phase II trial for idiopathic pulmonary fibrosis, rentosertib reportedly lowered proteomic biological age in the treated group—potentially signalling disease regression, systemic rejuvenation, or both. If confirmed, this could be a landmark advance in gerobiology. buff.ly/WSFNu7T

Geoffrey Hinton admitted he was wrong that AI would replace radiologists. Healthcare demand is elastic, so cheaper/faster scan reading creates more scans, and radiologists do more than interpret images. What many people fail to consider is AI automating tasks is not the same as eliminating the job

⚠️ A reminder that #CARTcells expansion beyond cancer comes with real safety challenges. Novartis has paused 8 trials of its CAR-T therapy in autoimmune diseases after 3 patients died following severe immune reactions. A major setback for an otherwise highly promising field. buff.ly/ApFP5Iy

His words came true. Democracy can hardly be expected to flourish in societies where political and economic power is being progressively concentrated and centralized. But the progress of technology has led and is still leading to just such a concentration and centralization of power. Aldous Huxley

Anti-CD19 #CARTCells can persist up to 10 years after infusion in patients with lymphoid malignancies. In one patient CART cells comprised 1.2% of circulating T cells at 9y. Long-term persisting CART cells were predominant double-negative (CD4−CD8−), effector-memory-like phenotype. buff.ly/FBYeuwR

Encouraging results for CAR-T in recurrent glioblastoma: intracranial B7-H3 CAR-T (TX103) was well tolerated in a Phase 1 trial (n=15), with no DLTs. Median OS reached 19.1 months, with 66.7% alive at 12 months and one sustained complete response. Phase 2 warranted. #CARTcells buff.ly/1hWb3c9

“You will get the country you voted for. That country is one where we will no longer innovate. We will be less secure, less healthy, and less wealthy because some community of people who influence what this country is do not see the value of science” - Neil deGrasse Tyson — It’s already too late.
