I can't tell you how excited I am to expand our synthetic/systems biology research into the world of proteins and, in particular, to the superfamily of G-protein coupled receptors (GPCRs). I am so grateful to all our wonderful collaborators and mentors (you know who you are!)

This vision grew out of the collective ideas, expertise, & creativity of a remarkable team of researchers who came together in our lab around a shared scientific challenge. I'm grateful for NIH's support, which gives us the opportunity to work together to tackle this challenge.

🌱🚨 Very proud and excited to share our PREPRINT, unveiling an advance for plant science, biotech, & synthetic biology in the first (of many) Khalil <> Gehring collaborations! www.biorxiv.org/content/10.6...

We synthesized these principles into a next-generation, precision plant transformation platform, yielding the T1 vector system — a platform that enables single-copy, backbone-free, readily mappable transgene integration with predictable expression.

We hope and expect this advance will greatly accelerate workflows for creating predictable, stable, and precisely engineered plants. Thanks for reading!

By screening ~1000s of transformants, we uncovered a previously under-appreciated role for the virulence-enhancing OVERDRIVE sequence as a key determinant of T-DNA copy number, insertion complexity, and vector backbone transfer.

We also found a fundamental tradeoff btwn transformation efficiency and integration quality. Thus, strategies focused solely on increasing transformation efficiency can compromise the quality of recovered transgenic lines.

We systematically evaluated multiple components of the T-DNA vector system at unprecedented scale, in relation to a comprehensive set of transformation outcomes. How? ...

Agrobacterium-mediated transformation underpins much of plant biology and biotechnology; however, it has long relied on highly inefficient screening to find the rare “good” transgenic lines hidden among many complex insertions -- one of the major bottlenecks for plant biotech.

Indeed, the T-DNA vector system has been widely used for decades. Despite this, the vector determinants governing transformation outcomes and quality remain poorly understood.

Led by the BRILLIANT @willshaw.bsky.social (pictured below), this work presents the T1 Vector System -- a next-generation, precision plant transformation platform that enables clean, single-copy, readily mappable transgene integration in Arabidopsis with predictable expression. Let’s dive in!

And here is another direct link to the preprint. Enjoy! www.biorxiv.org/content/10.6...

🚨 An exciting frontier is bringing synthetic biology to MULTIcellular systems. To effectively engineer them, we need design principles. What are the benefits of group formation? What are the costs? See our new preprint led by @heidiklumpe.bsky.social to learn more!👇 www.biorxiv.org/content/10.6...

The preprint from my work @mokhalil.bsky.social and Mary Dunlop's groups at BU is out on bioRxiv! As new tools come online to engineer multicellularity, we asked: how does sticking cells together into larger groups affect their fitness and function?
