I’m very happy to share that part of my postdoc work is now out in Science Advances! We find that a chimp-specific endogenous retrovirus (PTERV1) insertion silences LINC00662 and that silencing this lncRNA alters neuronal maturation in neural organoids 🧬🧠🐒 1/6 www.science.org/doi/10.1126/...

Together, these findings support a role for PTERV1-driven silencing of LINC00662 chimps. As LINC00662 seems to play an important role in human brain development, this retroviral insertion might habe contributed to the evolutionary divergence of human and chimp brains. 5/6

We silenced LINC00662 in human iPSC-derived neural organoids to test its role. This reduced TUJ1 staining in the neurite-rich intermediate zone at day 30, without changing organoid growth or cell-type composition. 3/6

Single-nucleus RNA-seq showed increased expression of gene linked to axonogenesis, axon extension and dendrite development in LINC00662-CRISPRi organoids. This may be a compensatory response to impaired neurite development. 4/6

We profiled DNA methylation at 158 PTERV1 insertions in chimpanzee stem cells and neural organoids. At an insertion within LINC00662, methylation spread into nearby DNA silencing LINC00662. Removing the PTERV1 from chimpanzee iPSCs restored LINC00662 expression. 2/6

How do pluripotent stem cells resist harmful interferon responses to safeguard development? Through total epigenetic lockdown of ligands, sensors and effectors of IFN-I. In our preprint, James Holt shares his PhD discoveries on the ground state of immune evasion 😊: www.biorxiv.org/cgi/content/...

We profiled the effects of our CRISPRi system targeting young L1 elements in human pluripotency & brain development. These elements deeply impact our transcriptome, but also have very distinct effects on their nearby genome. Check it out!! 🧬👾 #TEsky #CRISPR #L1 #development #epigenetics

Profiling and targeting young L1s is no easy matter 🧐 check out our new preprint to see how we delve into transcriptional and epigenomic profiling of L1s, and investigate their involvement in pluripotency and early brain development 🧠

Always exciting and rewarding to share what we do in the lab with the new generations. Hopefully we even managed to inspire some young brains! So happy to be able to participate in #Unistemday2024 🧠🧬🔬 @lundstem.bsky.social
