1/3 A few snapshots from the past intense days at the University of Helsinki for our FLIM and FCS course! We combined lectures, hands-on microscopy and analysis with plenty of questions and discussion. A fantastic group of researchers to learn with. 🔬 #FLIM #FCS #Microscopy

Spoiler: lots of 😍 SPY555-FastAct live cell movies in there.

Our June issue is here! rupress.org/jcb/issue/22... The cover shows a maximum intensity projection from a confocal image stack showing an angiogenic sprout emerging from an engineered microvessel. From @lakynm.bsky.social et al. of the Matthew Kutys lab: rupress.org/jcb/article/... #Cytoskeleton

And it's not all: For an exceptional new product, an exceptional new casing! We designed new boxes that we will gradually deploy to replace our old metal boxes. 4 designs will be randomly used for each product!

What a probe! Congratulations @cepourroy.bsky.social! We're happy to share that we re working hard on making CenSpark probes available to the centriole aficionados in the coming weeks!

Cédric Pourroy@cepourroy.bsky.social

CenSpark story is now published in @natchembio.nature.com. We engineered a dual-ligand fluorescent probe to selectively "light up" centrioles & cilia. Massive thank to Pierre Gönczy, @lreymond.bsky.social and @ghatzopoulos.bsky.social for the guidance. Check it out 👉: doi.org/10.1038/s415...

Organoids stained with Spirochrome probes for actin and DNA! 😍

Valentin Dunsing-Eichenauer@dunsingvalentin.bsky.social

Painted 100+ lung organoids for Easter, stained with live-cell Cell Tracker/ @spirochrome.com dyes, imaged at our @leicamicrosystems.bsky.social Viventis Deep light-sheet 🔬 at Charité (soon @Si-M - Der Simulierte Mensch) and visualized using inverted lookup tables designed by @christlet.bsky.social

Our paper is now out in Science! Super excited to share our discovery that #mitochondria #pearling is the elusive mechanism driving the regular distribution and inheritance of #mtDNA nucleoids 🧬 [1/6]

🤩 Another amazing example of U-ExM actin staining with HAK-actin by @dudinlab.bsky.social et. al.

At this high resolution, we also caught something new: Using the new #HAK-Actin #UExM probe, we saw that #Actin accumulates at the furrow base before compartments fully seal ➡️ they're still open at the bottom. ➡️ The #multicellular epithelium-like stage is shorter than we previously proposed.

An amazing example of actin imaging with our expansion microscopy probe for actin HAK-actin™. These are U-ExM expanded platelets stained with HAK-Actin (magenta bottom), (anti-integrin αIIb magenta top), and fibrin (fibrinogen-A488, green). Movie from preprint www.biorxiv.org/content/10.6... 🧪

🧵 1/5 A new BioRxiv preprint reveals that membrane tension acts as a mechanical gate for lung cell fate during development! 🫁 Our Flipper-TR probe was key to assess live Cell Membrane Tension (CMT) Here’s how Flipper-TR unlocked a hidden layer of lung development & fibrosis. 👇 #Mechanobiology

4/5 The "Stuck" State: Flipper-TR also exposed a fibrosis mechanism. In healthy lungs, tension drops to allow repair. But in fibrotic lungs (IPF), Flipper-TR showed tension stays high. This high tension state traps cells in a dysfunctional "transitional state," preventing healing.

2/5 The Challenge: Scientists knew various signals guided lung differentiation, but how do cells "know" when to specialize? Here comes Flipper-TR: A mechanosensitive dye that changes its fluorescence lifetime based on CMT. High tension = Long lifetime; Low tension = Short lifetime

3/5 Mapping the Invisible: Using Flipper-TR + FLIM imaging, researchers visualized tension in live lung tissue. They discovered a drop in membrane tension before cells differentiate into AT1 or AT2 alveolar cells. Even cooler? It mapped "emergent zones" of low tension where differentiation starts!
