To invade red blood cells, malaria parasites create a temporary molecular portal that lets it slip into the cell — and then disappears behind it. Researchers have finally detailed the proteins that let it break in without tripping the alarms. www.quantamagazine.org/malaria-para...

MemBrain v2 is on @natmethods.nature.com now? I never had a @biorxivpreprint.bsky.social with 164 citations before😅 ...Which is also to say: Thank you to the amazing #TeamTomo community, many of whom not only beta tested, but also contributed diverse training data. This one belongs to all of us! 🤗🔬🧪

A wild publishing ride to get here... but finally! (first bioRxiv: January 5th, 2024 🙃) Amazing work from @lorenzlamm.bsky.social and colleagues 🤗 Special thanks to the community who jumped early on the boat, sharing feedback and training data to make MemBrain v2 a reality 🙏🏽

How do African trypanosomes regulate the complement system? Check out our new study from @BiochemCook, in which he identified a new trypanosome receptor which binds to the core C3bBb convertase of the complement system and blocks its activity. Real all about it! doi.org/10.64898/202...

The full structure of the AMA1-RON complex sheds light on ongoing efforts to develop antimalarials targeting parasite invasion, and reveals the first clues into how the moving junction works, pointing toward a massive molecular machine that actively remodels the host cell membrane during invasion!⚙️🗜️

Finally, Messi teamed up with our newest student Daphne Kaxiras to explore a new avenue for antimalarial design. Using the structure as a guide, they used #BindCraft to design and validate a mini-protein binder that inhibits parasite invasion by blocking the formation of the moving junction! 🚫🦠

The moving junction only exists for the 60 sec it takes for a parasite to hijack a host cell, so Messi found a way to stall invading parasites on host cells and purified the native complex directly from invasion-stalled parasites for #Cryo-EM ❄️🔬The final structure looks a lot like a sailboat! ⛵

Thrilled to share the updated video of my paper in Nature with Maciej (under supervision of @simonemattei.bsky.social & @jomaalab.bsky.social) Proud to use Cryo-ET to uncover how hibernation factors are key to translation restart! To model PTMs in situ and render animations fulfilled my PhD dream!🥹🎉

Happy to share our preprint! Using massively parallel reporter assays, single-molecule imaging, deep learning and cryo-EM, we decode the sequence requirements for translation initiation and describe the “extended translation initiation sequence” (eTIS) that modulates start codon recognition. 🧵👇
