📣 Interested in non-coding disease-causing variants? Check out our review "Mechanisms underlying disease-causing variants in promoters and enhancers". Interesting mechanisms, challenges and future perspectives. Great to work with @wbickmor.bsky.social, Kun and Ryan! www.nature.com/articles/s41...

nature.comMechanisms underlying disease-causing variants in promoters and enhancers - Nature GeneticsThis Review discusses how rare-disease-causing variants in the noncoding genome impact gene regulation, why these examples are so few and how new approaches could accelerate discovery of noncoding var...

It was great working with students Kun and Ryan and @hannahlong.bsky.social, combing the literature to try and find examples of bona fide disease-causing variants in non-coding elements - promoters, enhancers and silencers. www.nature.com/articles/s41...

For the past 30 years, “whole-genome sequencing” has been a misnomer. Today the T2T Consortium publishes a dozen papers heralding a future of truly complete genomes for humans and nearly any vertebrate 👨‍🔬🐒🐦🐀🦒🐎🫏🐹🐟 (sorry, no salamanders): www.cell.com/consortium/t... 🧵[1/15]

A biochemical curiosity: it is much more straightforward to go enzymatically from 5mC > T than it is to go from 5mC > C

Happy to share our new review on X-chromosome reactivation in the germline, just published in @biochemsoc.bsky.social Transactions! We highlight the latest discoveries on how this epigenetic mechanism unfolds during mammalian germline reprogramming. Hope you enjoy the read 😊

portlandpress.comReprogramming the inactive X chromosome: dynamics and insights from the germlineAbstract. Germline reprogramming is an essential process that resets the epigenome prior to gamete formation. Primordial germ cells (PGCs), the progenitors of oocytes and spermatozoa, undergo extensiv...

Happy to share our latest paper. Our findings uncover a unique chromatin architecture and spatial chromosome arrangement in gonadal germ cells and document that alongside global DNA demethylation, the germline epigenetic reprogramming involves reorganisation of the 3D genome.

nature.comGlobal reorganization of genome architecture at the transition to gametogenesis - Nature Structural & Molecular BiologyHuang, Rigau and colleagues observe major changes in how DNA is organized in early germ cells before they start developing into sperm or eggs. These results show that germline removes structural ‘memo...

The 'Handler genome' of OSCs, a rare and stably growing cell line that runs a piRNA pathway to silence transposons. Dominik (@86dominik.bsky.social) assembled the genome of this Drosophila cell line. Besides making some cool findings, the goal was to turn this into useful resource for the field.

Dominik Handler@86dominik.bsky.social

When transposons jump, genomes diverge - even in cultured cells. I am happy to share our new preprint: a chromosome-scale genome assembly for Drosophila OSC cells, one of the key model systems in the piRNA field, especially for nuclear piRNA biology. 🧬🧵 (1/12)
